Can anyone tell me how far it is to ride from the N. Jefferson trailhead to Holt Summit. Also, is it a safe route, meaning not risking your life riding on a busy highway. :)

It is only four miles or so but I wouldn't recommend it. Uphill and on two-lane road with traffic flying by. There's a wide shoulder so it is generally "safe" though not pleasant. I've only done it once and would never do it again.
