Is there a bike route over the Missouri where I-64 crosses ? That is, can you leave the KT and go into Chesterfield without a major detour ?

Yes a brand new one with a pedestrian section, after you cross the bridge the bike path will split take the path to the right and it will take you to the Chesterfield Valley.
